The M current, IM, a voltage-dependent non-inactivating K+ current, was recorded in NG108-15 neuroblastoma × glioma hybrid cells, using the whole-cell mode of the patch-clamp technique. We studied the effect of arachidonic acid, other fatty acids and inhibitors of the arachidonic acid metabolism. In relatively high concentrations (25–50 M) arachidonic acid first increased and later decreased the current, Ih, which holds the membrane potential at –30 mV and mainly flows through open M channels. It shifted the midpoint potential, Vo, of the relation between M conductance, gM, and membrane potential, V, to more negative values and decreased the maximum conductance ¯gM and the time constant
M. In smaller concentrations (5–10 M) arachidonic acid merely decreased Ih and ¯gM with little effect on Vo and
M. Eicosatetraynoic acid and docosa-hexaenoic acid acted similarly to arachidonic acid whereas stearic acid had no effect. Of the three enzyme inhibitors studied, nordihydroguaiaretic acid acted similarly to arachidonic acid. i. e. caused a biphasic change in Ih. Indomethacin and quinacrine caused, respectively, a pure increase and a pure decrease of Ih and ¯gM. Possible explanations are build-up of internally produced arachidonic acid, depletion of eicosanoid products or an inhibitory effect unrelated to arachidonic acid metabolism. 相似文献
Summary Free cholesterol in lipids from the scalp and hair is predominantly a constituent of epidermal lipids. Therefore, a reduction in cholesterol content induced by a drug indicates a reduction in cell turnover in the epidermis. As, according to the literature, increased cell turnover in the epidermis results in formation of dandruff, a reduction in the proportion of cholesterol should indicate inhibition of the formation of dandruff. Conversely, an increase in free cholesterol should generally indicate a keratolytic effect. So unequivocal an interpretation has not so far been possible in persons with dandruff, as it was not known whether free cholesterol was increased or decreased. In addition, this interpretation was not possible after use of antimicrobial substances, as in vitro investigations had failed to exclude microbial esterification of cholesterol on the scalp. The present investigation has shown that correlation of free cholesterol level with cell turnover is permissible in patients with dandruff, even if antimicrobial drugs are being tested. 相似文献
We describe a rare case of medulloblastoma in an adult woman with histological findings suggesting an origin for this neoplasm
in the external granular layer or its remnants. The patient presented with cerebellar dysfunction, and neuroimaging revealed
a right cerebellar mass lesion. Pathological examination of the operative specimen revealed a medulloblastoma with occasional
areas of neuronal or glial differentiation. Zic protein was also detected immunohistochemically in the tumor cells. The tumor
cells were mainly distributed in the subarachnoid space and extended to the cerebellar parenchyma through the perivascular
space to form tumor nodules. A suggestive finding, as concerns the origin of this neoplasm, was that the tumor cells were
also spread evenly along the subpial zone of the molecular layer, reminiscent of the cellular architecture of the fetal external
